Abstract: Lipoapoptosis is a fatty acid-induced apoptosis that occurs in obesity and ageing. However, the molecular basis of lipoapoptosis is still unclear. Using Schizosaccharomyces pombe as a simple model for lipoapoptosis, we examined the roles of selected putative mediator proteins Bzz1p, Pck1p, Pck2p, Rad9p and Apg6p in lipoapoptosis. In this study, we found that Bzz1p, Pck1p and Rad9p might be involved in the activation of lipoapoptosis in DKO (a??dga1 a??plh1) cells, but not Pck2p and Apg6p. We also demonstrated that rapamycin resulted in enhanced ROS production and necrotic cell death in logarithmic phase DKO cells, with dying cells lacking both DNA and nuclear fragmentation. Lastly, we showed that mitochondria did not participate in lipoapoptosis of DKO cells and therefore suggest that lipoapoptosis is regulated by a molecular mechanism distinct from that of conventional metazoan apoptosis.
