Abstract: Reports from different continents reported that Enterococcus faecalis this bacterium was associated with failed root canal treatment. It was hypothesised that chemo-mechanical preparation may increase the propensity for this bacterium to adhere and survive within roots.Irrigants used during root canal treatment were used to treat type I collagen membranes and examined for denaturation, using Circular Dichroism and Differential Scanning Calorimetry. Bacteria adhesion assays to treated collagen were assessed using Confocal Laser Scanning Microscopy. Adhesion force of E. faecalis to collagen was assessed using Optical Tweezers. Chemical changes to dentine were monitored using Zeta potential measurement and Fourier Transform Infra-red Spectroscopy and bacteria adhesion assay on dentine was assessed using fluorescent microscopy and adhesion force assessed by Atomic Force Microscopy. All chemicals used denatured collagen and altered dentine, and had an impact on the bacterial adhesion assays and different sequences led to an increase in E. faecalis adhesion.
