Abstract: Filopodia are cellular protrusions crucial for axon guidance, embryonic development, and wound healing. The exact mechanism(s) by which they arise are not fully known. In this study, the Rho GTPase Rif and Diaphanous-related formin mDia2 were shown by time-lapse imaging of live neuroblastoma cells to mediate an independent pathway for filopodium formation that does not involve effectors of the Cdc42-driven pathway, namely N-WASP, IRSp53, and Mena. Twenty-nine novel putative Cdc42 interacting proteins were picked up by yeast two-hybrid screens, of which at least three (FHOD1, RanBP9 and APC) lack known Cdc42-binding motifs. Also a Diaphanous-related formin, FHOD1 formed stress fibres rather than filopodia. In addition, at least 13 potential Rif interactors were identified by mass spectrometry analysis of tandem affinity purified protein complexes. Further work to validate their interactions with Rif and explore their possible cytoskeletal roles will lead to a fuller understanding of how filopodia form.
