Abstract: Previous studies have strongly implicated the loss of functional RUNX3 to be causal to the genesis and progression of gastric cancer. Despite its importance, much remain to be learnt of RUNX3's molecular functions in this tissue. This study aims to understand the role of RUNX3 in gastric homeostasis through the identification of direct RUNX3 target genes. This was achieved by a series of expression microarray analyses in which RUNX3 was transiently re-introduced into a RUNX3-negative gastric epithelial cell line, AGS. Subsequently, fifteen candidate target genes were selected and validated across a panel of gastric epithelial lines. S100A2 was induced by RUNX3 in all cell lines tested. In line with the notion that S100A2 is a RUNX3 target gene, RUNX3 transcriptionally activated S100A2 in a DNA-binding dependent manner. Additionally, clinical relevance of this work was demonstrated by a positive correlation of the expressions of RUNX3 and S100A2 in human gastric samples.
