Abstract: Hypericin (HY), a natural photosensitizer, is able to detect bladder cancer significantly better than white light endoscopy. However, its lipophilicity complicates the administration of HY for clinical applications. The delivery of HY was optimized using a novel biocompatible solvent and penetration enhancer, N-methyl pyrrolidone (NMP) for better therapeutic outcomes. The safety of NMP in human bladder carcinoma MGH cells and chick chorioallantoic membrane (CAM) were evaluated. Delivery of HY to MGH cells and CAM were performed. 3D confocal microscopy was used to report real-time sub-cellular localization of HY into cells. Photobleaching kinetics of HY and fluorescence enhancement properties of these formulations were defined by spectroscopy. Finally, the HY-NMP formulations were evaluated in CAM implanted with MGH cells. HY-NMP formulations were significantly better than the plasma protein-based formulations by producing higher contrast between tumor and adjacent tissues within 1 h. This study demonstrated the potential of CAM for studying drug transport.
