dc.description.abstract | In this research, a novel approach to tissue engineer skin was developed, to alleviate problems of graft contraction in current techniques. A weft-knitted poly(lactic-co-glycolic acid) mesh with mechanical properties comparable to human skin was fabricated as the matrix for dermal regeneration. The mesh was capable of supporting the attachment and proliferation of human dermal fibroblasts, and evoked minimal host tissue response in vivo. Culturing the mesh with fibroblasts sheets produced three-dimensional, collagen-rich dermal constructs which did not contract in vitro, and supported the regeneration of a premature epidermis. The constructs were viable, metabolically active, secreted growth factors and registered a take rate of 100% when transplanted onto full-thickness wounds in rats. The extent of contraction of wounds grafted with the constructs was comparable to autografts. This technique offers an alternative approach for tissue engineering a skin equivalent with mature extracellular matrix and reduced contraction both in vitro and in vivo. | |
