Abstract: Biomatrices for human tissue regeneration will change upon irradiation and extended storage. This study, using a human acellular dermal matrix (ADM), investigated the effects of radiation-induced free radical formation and storage stability during accelerated aging conditions. ADM was irradiated with gamma ray or stored at different relative humidities. Changes in ADM properties and their effect on cell proliferation were studied. Free radicals in ADM were studied using electron paramagnetic resonance spectroscopy; structural modifications were evaluated with scanning electron microscopy; mechanical properties were examined with Instron tester; and cell proliferation was assessed through growth and differentiation of fibroblasts. Treated ADM showed a more condensed and organized collagen fiber network and greater mechanical strength. Both treatments had enhanced the expression of fibroblast collagen type I and matrix degradation. It is concluded that structural modification in ADM by gamma irradiation and accelerated aging treatment influenced in vitro cellular behavior and matrix remodeling.
