Abstract: Zebrafish is a good vertebrate model for the study of early development. We took advantage of this model organism and employed different strategies in the attempt to elicit novel genes involved in hematopoietic program. Firstly, forward genetic screening was carried out to search for T-cell early development and thymus organogenesis deficient mutants by using rag-1 as a marker. Of the 600 mutagenized genome we screened, 13 mutants were recovered. Secondly, Affymetrix arrays were applied to analyze zebrafish cloche mutant, which has defects in hematopoietic and vascular lineages. 18 uncharacterized zebrafish genes were found to be down-regulated in cloche mutant. 8 of them were predominantly expressed in hematopoietic and vascular endothelial cells. Different expression patterns of these genes indicated their diverse functions during hematopoiesis.
