Abstract: Day 8 prednisolone response is highly predictive of outcome for childhood ALL. We studied the paired gene expression profiles of leukemic cell lines before and after exposure to prednisolone to define the genes responsible for this prognosis. Five genes (six probe sets) were consistently elevated post prednisolone among the three sensitive cell lines when compared to their controls and the resistant line. This trend was confirmed using real-time quantitative PCR. This pattern may be a useful signature for predicting response to prednisolone in prospective evaluation in our current leukaemia trial. Gene expression profiling may provide important clues for management of childhood ALL.
