dc.description.abstract | Sulfatide, the natural sulfated derivative of galactosylceramide, is known to bind to several extracellular matrix (ECM) glycoproteins including tenascin-C (TN-C), whose expression is highly up-regulated in many tumors such as glioma. It is described here a sulfatide-containing liposome which targets to the glioma cells. In vitro experiments with TN-C expressing glioma cell lines demonstrated that robust cellular uptake and ECM binding of the liposomes depended specifically on the presence of sulfatide as the liposomal component. Following binding to the ECM, the sulfatide containing liposomes were internalized via both caveolae- and clathrin-dependent mechanisms, with significant amount being largely intact in the cytoplasm after a 5-h observation period. It has also been examined that TN-C was involved in binding of sulfatide-containing liposomes to the ECM. Taken together, it suggests that sulfatide-tenascin interaction may lead the sulfatide-containing liposome to target the ECM of glioma cells. These findings demonstrate the potential application of this drug delivery system in delivery of chemotherapeutics such as doxorubicin, which should improve the targeting specificity and treatment effectiveness of the latter. | |
