ROLE OF THEMIS IN TYPE 2 DIABETES. ScholarBank@NUS Repository. | Abstract: | Type 2 diabetes mellitus (T2D) is a metabolic disorder that results in high blood glucose due to insulin resistance. There has been increasing evidence of involvement of the immune cells in development of T2D. Themis is a T cell specific protein, which has been shown to set the threshold for positive and negative selection in the thymus. T cells from Themis-/- mice show defects in the TCR stimulation-induced glucose uptake. Themis-/- mice tend to develop diabetic phenotype as they age, which suggests a role of Themis in T2D. Data from Themis conditional knockout mice and T cell depletion experiments proves that this is a T cell intrinsic phenotype. Preliminary data from conventionalizing germ-free mice with microbiome from Themis-/- and Themis+/+ mice suggests regulation of T cell-microbiome axis by Themis in metabolic disorder. We have also seen differences in TCR repertoire of Themis-/- mice. These results would be discussed in relation to the development of T2D. | URI: | http://scholarbank.nus.edu.sg/handle/10635/149548 |
