Abstract: Wild type Adenovirus (Ads) was purified and could be easily labelled with fluorescence and there was no significant difference in infectivity between fluorescence-labelled Ads and original ones. Synchronization of fluorescence-labelled Ads could be achieved readily through temperature control. Human and mouse centrin was cloned and expressed as a chimerical protein with GFP that served as a location marker. Ads escaped from endosome were purified and monoclonal antibodies against the escaped virus were developed, this antibody can be used to study the assembly of host transportation machinery. Such a system can be used as a powerful tool in the study of virus interaction with host transportation machinery. In addition, study of differences in transportation pattern between Ad and non-viral gene delivery system (PEI) showed that the transportation efficiency of Ad is much higher than that of PEI.
