Abstract: Acute Graft-versus-host disease (aGVHD) is initiated by allogeneic T cells which preferentially infiltrate and damage certain host organs. Our study observed a distinct pattern of chemokine expression in these target organs which might be responsible in directing the infiltration of alloreactive T cells. Proliferating and non-proliferating allogeneic T cells were isolated from lymphoid organs of diseased mice, and both populations were able to induce aGVHD in secondary recipient mice. Characterization on these two populations of cells gave interesting results. Amelioration of aGVHD was attempted by administration of an immunotoxin gene (DT390-IL-2) into the diseased mice, and the results are encouraging.
