Abstract: ORCHIDS ARE MEMBER OF THE FAMILY ORCHIDACEAE, WHICH IS ONE OF THE LARGEST FAMILIES OF FLOWERING PLANTS. INVESTIGATION OF FLOWERING MECHANISMS IN ORCHIDS CONTRIBUTES TO UNDERSTANDING OF PLANT REPRODUCTIVE BIOLOGY AND ORCHID BREEDING. IN THIS STUDY, WE CHARACTERIZED DOFT AND DOFTIP1, BOTH OF WHICH ARE INVOLVED IN REGULATION OF THE FLORAL TRANSITION IN DENDROBIUM CHAO PRAYA SMILE. IN ADDITION, OUR RESULTS DEMONSTRATED THAT DOFT ALSO PARTICIPATED IN PSEUDOBULB FORMATION AND FLOWER DEVELOPMENT. FURTHERMORE, THE EARLY FLOWERING CAUSED BY OVEREXPRESSION OF DOFT UNDER THE PHLOEM-SPECIFIC SUC2 PROMOTER IN ARABIDOPSIS SUGGESTING THAT DOFT MAY ALSO ACT AS A MOBILE FLORAL SIGNAL IN ORCHIDS. MOREOVER, INTERACTION BETWEEN DOFT AND DOFTIP1 PROVIDES EVIDENCE THAT DOFTIP1 MAY PARTICIPATE IN DOFT TRANSPORT IN ORCHIDS. THIS STUDY PROVIDES NEW INSIGHTS INTO MOLECULAR REGULATION OF FLORAL TRANSITION IN ORCHID, AND ENGINEERED TRANSGENIC ORCHIDS WITH EARLY FLOWERING PHENOTYPES COULD SERVE AS NOVEL BREEDING M
