Abstract: THE MITOTIC SPINDLE IS A DYNAMIC STRUCTURE WHOSE MOVEMENTS ARE CAREFULLY REGULATED DURING THE CELL CYCLE. SUCH REGULATION IS NECESSARY TO ENSURE PROPER SEGREGATION OF CHROMOSOMES TO THE PROGENY. KIP2P IS A MOTOR PROTEIN THAT REGULATES CYTOPLASMIC MICROTUBULE STABILITY AND HELPS IN SPINDLE POSITIONING. IT IS KNOWN THAT KIP2P LEVELS ARE REGULATED DURING THE CELL CYCLE, HOWEVER THE PURPOSE OF THIS REGULATION REMAINS UNEXPLORED. IN THIS STUDY, WE SHOW THAT KIP2P LEVELS MUST BE REGULATED TO ENSURE TIMELY CHROMOSOME CAPTURE. KIP2P IS NECESSARY DURING EARLY MITOSIS TO STABILIZE SPINDLE ORIENTATION, AS THE ABSENCE OF KIP2P LEADS TO TUMBLING OF SPINDLES, WHICH IN TURN LEADS TO A DELAY IN CHROMOSOME SEGREGATION. WE ALSO EXPLORED THE POSSIBLE MECHANISMS OF KIP2P REGULATION DURING THE CELL CYCLE. WE FOUND THAT CDK1/CLB5 PHOSPHORYLATION MAY BE IMPORTANT FOR MAINTAINING KIP2P LEVELS AND CDH1 MEDIATED DEGRADATION MIGHT REGULATE KIP2P LEVELS DURING LATE MITOSIS AND G1 PHASE.
