dc.description.abstract | Evidences have confirmed that activated microglial cells with high migratory and cytotoxic properties were responsible for the neurodegeneration of injured neurons in the central nervous system (CNS). Recently, it has been reported that an extracellular matrix protein, Tenascin-R (TN-R), was anti-adhesive to activated microglia, thus may play a role in neuronal protection against the neurodegeneration after CNS injury. To find out which domain of TN-R is responsible for the anti-adhesive effect to the microglia, distinct domains of TN-R were bacterially expressed as GST-fusion proteins, and repulsion assay was performed with the primary microglia culture. Optic nerve transection model was used as an example of CNS injury. Effective domain of protein was given intraocularly after the operation with various concentrations. Retinal microglia cells were analysed by OX42 immunohistochemistry at various survival intervals. Both in vitro and in vivo studies demonstrated that the epidermal growth factor-like (EGF-L) domain of TN-R has anti-adhesive effect on activated microglia compared to GST control groups. | |
