dc.description.abstract | Membrane trafficking is an important cellular process whereby proteins are sorted and transported to various compartments within the cell. The Golgi-apparatus plays a central role in the regulation of membrane trafficking and signaling through novel Golgi-localised G-protein coupled receptors (GPCRs). Through data mining, we identified GPR108 as a novel Golgi-localised GPCR with a potential role in the membrane trafficking pathway. This study reports an in-depth characterisation of GPR108, showing that GPR108 is likely a seven-span transmembrane protein, with a cytoplasmic C-terminus which is responsible for its Golgi-targeting by aiding its endosome-to-trans-golgi-network (TGN) trafficking. Furthermore, functional analysis revealed that knockout of GPR108 does not affect Golgi structure and morphology, endosomal distribution pattern or other aspects of membrane trafficking. However, I find that GPR108 modulates the levels of c-fos and phosphorylated forms of EGFR and STAT3 upon stimulation with complete media. These results imply a potential role for GPR108 in the regulation of EGFR-STAT3 signaling. | |
