Abstract: CD8 T cells play a crucial role in providing protective immunity against a wide range of pathogens and because one of their primary functions involves the killing of other cells, their activation is tightly regulated. Dendritic cells (DCs) regulate the activation and differentiation of CD8 T cells through a variety of signaling pathways mediated by costimulatory molecules expressed on both cell types. One major costimulatory molecule is CD40L and although CD40L expression was conventionally associated with CD4 T cells, we studied the expression of CD40L on CD8 T cells and the role it plays in CD8 T cell-DC interactions. We generated and characterized CD40L-expressing CD8 T cells both in vitro and in vivo and found that CD40L is highly expressed by a subset of CD8 T cells. We also investigated the existence of a CD40L-dependent mechanism where CD8 T cells and DCs cooperate to maximize CD8 T cell responses.
