ScholarBank@NUS Repository. | Abstract: | Human hepatocellular carcinoma HepG2 cells that stably overexpressing human PlGF1 and PlGF2 were generated by transfection and introduced subcutaneously into immunodeficient mice to study the effect of PlGF during tumor development. Overexpression of PlGF1 in HepG2 promoted modest but significant tumor growth and tumor angiogenesis without affecting VEGF homodimer level. The increased tumor angiogenesis was associated with upregulated levels of MCP-1 and active MMP-9 which resulted from stimulation of macrophage infiltration via activation of VEGFR1 by PlGF1 overexpression. In contrast, overexpression of PlGF2 in HepG2 reduced VEGF homodimer availability by formation of PlGF2/VEGF heterodimer, which resulted in downregulated tumor angiogenesis and increased tumor apoptosis with increased TNF-I? level. Collectively, cooperative action of PlGF and VEGF together with tumor microenvironment is important in HepG2 tumorigenesis. 20 percent reduction of PlGF production in human placenta choriocarcinoma JAR tumors by antisense PlGF expression failed to influence tumor growth with unperturbed VEGF homodimer and VEGF/PlGF heterodimer levels. | URI: | http://scholarbank.nus.edu.sg/handle/10635/14257 |
