dc.description.abstract | In personalized medicine, it is essential to develop biosensing systems that can regenerate the sensor probes for continuous dynamic monitoring of disease-related biomarkers or therapeutics. This thesis presents an electrokinetics-integrated electrochemical aptamer-based sensor (e2-aptasensor) for both continuous monitoring of biomolecules and online regeneration of the sensor probes in biological samples. The electrokinetic concentration, also known for its underlying mechanism ion concentration polarization (ICP), works synergistically with e-aptasensors to achieve buffer-free regeneration with minimal disturbance. We first performed basic characterization of the e-aptasensors and the ICP operation parameters, and then we conducted in vitro testing of ATP in synthetic saliva and DOX in human serum. Results showed that the e2-aptasensors could perform continuous dynamic monitoring in the biofluids for hours. Given the e2-aptasensors’ relative simple operation and all-electronic architecture, it opens the possibility in developing such device into a portable system for personalized health monitoring or point-of-care diagnosis. | |
