ScholarBank@NUS Repository. | Abstract: | This thesis discusses the development of a novel nanodevice comprised of gold nanorods (NRs) with a human serum (HS) protein corona loaded with Chlorin e6 (Ce6) for simultaneous photothermal (PTT) and photodynamic therapy (PDT). In vitro uptake of Ce6 by Cal 27 oral squamous cell carcinoma (OSCC) cells was increased 57-fold with NR-HS-Ce6 compared to free Ce6. Combined PTT+PDT achieved 95.2% cell kill with only 50pM NR-HS-Ce6. Substituting the HS corona with mouse serum (MS), in vivo intravenous delivery of NR-MS-Ce6 into tumor-bearing nude mice resulted in peak tumor accumulation of 560.3µg Au/kg tissue. Combined PDT+PTT by NR-MS-Ce6 achieved complete tumor regression within 19 days. The success with NR-HS-Ce6 was further expanded upon to co-load Doxorubin (Dox) with Ce6 onto NRs, resulting in simultaneous PTT, PDT and chemotherapy (CTX) achieving 98.7% cell kill with 15pM NR-HS-Ce6-Dox.
