Abstract: In this study, characterization and localization of CD38 in murine testes were carried out. For the first time we report the presence of functionally active CD38 in murine testes through Western-blotting, immuno-histochemical staining and ADP-ribocyl cyclase assay. Through affinity chromatography and deglycosylation study, we also showed a differential post-translational glycosylation pattern between testis CD38 and liver CD38. Further investigation showed the specific localization of CD38 in the spermatid acrosome. Mapping of the expression of CD38 during spermiogenesis revealed its expression only in the Cap phase and Acrosome phase. Our study on CD38-/- mice testes showed significant levels of ADP-ribosyl cyclase compared to wild type mice, implying a functionally active ADP-ribosyl cyclase separate and distinct from CD38 was upregulated in testis when CD38 was genetically knocked out. Taken together, our results suggested a previously unknown role for CD38 and the associated cADPR-mediated Ca2+ signaling pathways in acrosome biogenesis during spermiogenesis.
