Abstract: Cervical carcinomas cause ~250,000 deaths per year worldwide, virtually all contain human papillomavirus DNA with 60% of them identified as HPV16. One of the two main oncoproteins is a??early protein 6a?? (E6) which targets p53 for degradation. E6 is expressed throughout cancer progression, necessary for the survival of the cancer cell and thus makes for an excellent drug target. Here, I present a novel way for the preparation and stabilization of high concentrations of monomeric E6, a prerequisite for structural studies, and evidence for the inherently flexible disposition of the E6 protein reflected in its large number of interaction partners, of which CBP/p300 is introduced here.
