dc.description.abstract | Purpose. To evaluate the efficacy of in vitro unaided transscleral penetration of Ganciclovir in the pig eye. Methods. Freshly enucleated whole adult pig eyes were immersed in Ganciclovir solutions of 20, 50 and 200 mg/ml for 2, 4 and 8 hours at 37 °C. Duplicates were used for each concentration level and time interval (total of 18 eyes). Vitreous was obtained and Ganciclovir concentrations were measured by using high performance liquid chromatography. One fresh cadaveric human eye was also studied using a 30 mg/ml bathing solution and 4 hours of immersion. Results. At 2, 4 and 8 hours of immersion, the average vitreal Ganciclovir concentrations in the pig eyes were 0.9, 1.9 and 5.6 mg/ml respectively with the 20 mg/ml solution; 2.7, 4.1 and 11.0 mg/ml with the 50 mg/ml solution; 2 9, 9 9 and 35.3 mg/ml with the 200 mg/ml solution For the human eye, the vitreous level was 2.3 mg/ml after 4 hours Multiple regression analysis was performed on data for the pig eyes.
