dc.description.abstract | Collagen I serves as an organic scaffold in bone and it has been reported that denatured collagen appeared to preserve the osteogenic differentiation potential of mesenchymal stem cells. This study investigated the effect of collagen I and gelatin on the attachment and mineralization of mouse calvaria pre-osteoblasts. Atomic force microscopy (AFM) imaging revealed that the collagen I matrix prepared was D-periodic, while the gelatin obtained upon denaturation exhibited decreased structural integrity. After a contact time of 3 minutes, AFM single-cell force spectroscopy showed that the force required to detach a pre-osteoblast from gelatin was significantly higher than that from collagen I. This is consistent with the finding that more focal adhesions were observed in cells which were plated on gelatin. On gelatin, the pre-osteoblasts also exhibited a higher extent of mineralization upon osteogenic induction. Therefore, compared to collagen I, gelatin appeared to enhance the attachment and subsequent mineralization of pre-osteoblasts. | |
