dc.description.abstract | (-)-Epigallocatechin-3-O-gallate (EGCG), a major ingredient of green tea, is recognized for its binding property and various beneficial effects, including anti-cancer effect. In this thesis, we have developed EGCG-based micellar nanocomplexes (MNCs) constructed by self-assembly of EGCG conjugated polyethylene glycol (PEG-EGCG) and small molecule chemotherapeutic, Sunitinib (SU). The SU-loaded PEG-EGCG MNCs (SU/PEG-EGCG MNCs) were obtained by the interaction between EGCG moieties and SU forming the core surrounded by PEG shell. The SU/PEG-EGCG MNCs efficiently suppressed tumor growth with higher tumor selectivity and longer blood half-life than intravenously injected free SU. This anti-cancer effect of SU/PEG-EGCG MNC was achieved by a much smaller amount of SU with no toxicity, as compared to conventional oral administration. The EGCG-based MNCs offered improved SU efficacy through highly specific tumor targeting, prolonged blood half-life and combinational effect between drug and carrier. This would overcome problems associated with conventional anticancer drugs, which usually have a narrow therapeutic window and severe side effects. | |
