Title: MAPPING QTL FOR RESISTANCE TO VNN DISEASE AND ANALYSIS OF TRANSCRIPTOME IN RESPONSE TO NNV INFECTION IN ASIAN SEABASS

Abstract: To understand the genetic architecture for resistance to VNN disease and investigate transriptome profiling in response to NNV infection in Asian seabass, I conducted QTL mapping using genetic maps consisting of microsatellites and SNPs and analyzed transcriptome profiling in response to NNV infection in Asian seabass. Primary QTL mapping detected thirteen QTL for resistance and ten QTL for survival time. I further constructed a high-density map consisting of 3000 markers generated by genotyping-by-sequencing. QTL fine mapping narrowed down one QTL to 1.76 cM, leading to identify pcdhac2 as the possible candidate gene. In addition, mRNA of Asian seabass epithelial cells challenged by NNV was sequenced by RNA-seq. A total of 385 million clean reads was obtained and de novo assembled into a transcriptome consisting of 89026 transcripts. Further transcriptome profiling analysis identified 251 differentially expressed genes, including rtp3, viperin, irf3, in response to NNV infection in Asian seabass.
