Abstract: Through acetone-butanol-ethanol (ABE) fermentation, Clostridium beijerinckii G117 produces butanol and butyric acid, which can then be esterified to butyl butyrate. Strain G117 also produces natively a small xylanase enzyme (22.6 kDa) which hydrolyzes xylan into fermentable sugars. To produce butyl butyrate without recovering butanol and butyric acid from culture media, organic solvent and Nafion are added to increase the esterification yield, but the toxic solvent is in direct contact with the bacterial cells. To improve the system, biphasic alginate beads containing an organic phase and lipase enzyme are developed. Apart from catalyzing esterification (31% yield), they also do not affect the growth of strain G117. Finally, a triphasic esterification system is developed in which the ester forms distinct oil droplets which facilitate the recovery of butyl butyrate. Multiple cycles of esterification (with product removal) improve the yield to 86%, which is the highest yield reported in aqueous solutions.
