Abstract: NON-CODING RNAS (NCRNAS) REGULATE GENE EXPRESSION IN MANY PROCESSES. GENOME-WIDE STUDIES IN MULTIPLE ORGANISMS IDENTIFIED THOUSANDS OF LONG NCRNAS BUT THE FUNCTIONS ARE NOT WELL-UNDERSTOOD. USING C. ELEGANS, WE INVESTIGATED THE FUNCTION OF 18 PREDICTED NCRNA GENES CLUSTERED NEAR BED-3, WHICH ENCODES A TRANSCRIPTION FACTOR REGULATING LARVAL MOULTS AND VULVAL CELL DIVISION. THE PRESENCE OF RNA TRANSCRIPTS WAS CONFIRMED FOR THREE NCRNA GENES, F25H8.13, F25H8.16 AND F25H8.21. WE FOUND THAT F25H8.16P::GFP IS EXPRESSED IN VULVAL MUSCLES, WHICH IS REQUIRED FOR EGG-LAYING. THE DELETION OF MULTIPLE NCRNA GENES, INCLUDING F25H8.16, CAUSED DEFECTS IN VULVAL MUSCLES, EGG-LAYING AND MALE TAIL MORPHOLOGY. THESE DEFECTS ARE ALSO FOUND IN BED-3 MUTANTS, SUGGESTING THAT THE DELETION AFFECTS BED-3 EXPRESSION. FLUORESCENCE IN SITU HYBRIDISATION CONFIRMED THAT THE DELETION REDUCED BED-3 EXPRESSION SPECIFICALLY IN SEX MYOBLAST CELLS, THE PRECURSOR OF VULVAL MUSCLES. ADDITIONALLY, THE PREDICTED NCRNA GENE C
