Abstract: CO-CULTURING PRIMARY HEPATOCYTE WITH NON-PARENCHYMAL CELLS (NPCS) IS GARNERING INCREASING INTEREST FOR DRUG TESTING APPLICATIONS IN VITRO. MAINTENANCE AND ENHANCEMENT OF LIVER SPECIFIC FUNCTIONS HAS BEEN ACHIEVED BY CO-CULTURING HEPATOCYTES WITH NON-PARENCHYMAL CELLS SUCH AS NIH-3T3 AND PRIMARY KUPFFER CELLS IN VARIOUS ENGINEERED PLATFORMS. HOWEVER, THERE IS LIMITED UNDERSTANDING IN THE UTILITY OF THESE PLATFORMS FOR DRUG INDUCED CYTOCHROME P450 (CYP) INDUCTION AND INFLAMMATION MEDIATED HEPATOTOXICITY. IN THIS THESIS, WE INVESTIGATED THE POTENTIAL APPLICATIONS OF RAT HEPATOCYTE/NIH-3T3 AND RAT HEPATOCYTE/KUPFFER CELLS CO-CULTURE FOR THESE APPLICATIONS RESPECTIVELY. THE HEPATOCYTE/NIH-3T3 CO-CULTURES IN RANDOM AND MICRO-PATTERNED CO-CULTURE CONFIGURATIONS DEMONSTRATED REPRESSED CYP INDUCTION COMPARED TO HEPATOCYTE MONO-CULTURES. WE FOUND THAT THIS PHENOMENON IS MEDIATED BY THE SOLUBLE FACTOR TRANSFORMING GROWTH FACTOR BETA 1 (TGF?1) AND NOT CONTROLLED BY HETEROTYPIC OR HOMOTYPIC INTERAC
