ScholarBank@NUS Repository. | Abstract: | LOWER RESPIRATORY INFECTIONS ARE THE FOURTH LEADING CAUSE OF DEATH WORLDWIDE, AND THE FIRST IN DEVELOPING COUNTRIES. AS ONE OF THE MAJOR RESPIRATORY INFECTIONS, INFLUENZA CAUSES MILLIONS OF DEATH EVERY YEAR, AND THE ASSOCIATED SECONDARY BACTERIAL INFECTION CONTRIBUTES A LARGE PORTION OF THIS NUMBER. THIS THESIS DEMONSTRATES THE IMPORTANT ROLE ANGPTL4 PLAYS IN LUNG INFECTION AND DAMAGE CAUSED BY INFLUENZA AND SECONDARY BACTERIAL INFECTIONS. ANGPTL4 WAS UPREGULATED THROUGH A STAT3-MEDIATED PATHWAY AND COLOCALIZES WITH LUNG TISSUE DAMAGE. TREATMENT WITH AN ANTI-CANGPTL4 MONOCLONAL ANTIBODY SIGNIFICANTLY REDUCED TISSUE LEAKAGE AND IMPROVED LUNG TISSUE INTEGRITY IN MICE. THESE RESULTS MAY FACILITATE NEW THERAPEUTIC STRATEGIES FOR THE TREATMENT OF THESE MALADIES. ACUTE LUNG DAMAGE INDUCED BY INFECTION IS A VERY COMMON OUTCOME FOR A VARIETY OF RESPIRATORY PATHOGENS AND OTHER DISEASES.
