THE TEAR FILM IS AN ACCESSIBLE SOURCE IN STUDYING OCULAR SURFACE DISORDERS AS IT IS DIRECTLY ASSOCIATED WITH THE CORNEAL SURFACE. INFORMATION-DEPENDENT-ACQUISITION LC-MS/MS AND ISOTOPE PATTERN MATCHED PEAK MINING ENABLED THE IDENTIFICATION OF 60 CHEMICALLY-DIVERSE METABOLITES IN HUMAN TEARS, OF WHICH 44 WERE REPORTED FOR THE FIRST TIME. GLOBAL METABONOMIC PROFILING WAS PERFORMED ON AN IN VITRO MODEL DRY EYE DISEASE. GYICEROPHOSPHOCHOLINE WAS IDENTIFIED AS THE PREFERRED OSMOPROTECTANT. INCREASED SYNTHESIS OF UDP-N-ACETYLGLUCOSAMINE AND ACTIVATION OF THE HEXOSAMINE PATHWAY UNDER HYPEROSMOTIC CONDITIONS WAS ALSO CONFIRMED WITH PROTEOMIC ANALYSIS. DIABETIC RETINOPATHY (DR) IS THE MOST COMMON MICROVASCULAR COMPLICATION OF DIABETES.
