dc.description.abstract | C-JUN, A MAJOR COMPONENT OF AP-1 TRANSCRIPTION FACTOR COMPLEX, IS INVOLVED IN A WIDE RANGE OF PHYSIOLOGICAL AND PATHOLOGICAL PROCESSES. HOWEVER THE GENE TARGETS THAT MEDIATE THESE PROCESSES REMAIN TO BE INVESTIGATED. ANALYSIS OF NOVEL C-JUN TARGETS BY WHOLE GENOME EXPRESSION PROFILES FROM EMBRYOS AND STRESSED PRIMARY MOUSE EMBRYONIC FIBROBLASTS (MEFS) SHOWED A SUBSTANTIAL NUMBER OF DIFFERENTIALLY EXPRESSED GENES BETWEEN C-JUN+/+ AND C-JUN-/- , WHILE ONLY A SMALL NUMBER OF GENES DIFFERED BETWEEN C-JUN+/+ AND C-JUNAA/AA. THUS C-JUN PHOSPHORYLATION IS REQUIRED ONLY FOR A SMALL SUBSET OF TARGET GENE TRANSCRIPTION. TO ELUCIDATE C-JUN¿S ROLE IN HEPATIC FIBROSIS (IDENTIFIED AS THE TOPMOST PATHWAY REGULATED BY C-JUN), WE UTILIZED C-JUN CONDITIONAL KNOCKOUT MICE TO INACTIVATE C-JUN IN (1) ADULT LIVER HEPATIC STELLATE CELLS (HSCS) AND (2) BOTH HEPATOCYTES AND HEMATOPOIETIC CELLS, BY USING COL-CREER AND MX-CRE TRANSGENIC MICE RESPECTIVELY.
