Abstract: EPIGENETICAL MODIFICATIONS HAVE BEEN IMPLICATED IN MAINTAINING CHROMATIN STRUCTURE, REGULATING TRANSCRIPTOME AND DICTATING THE BIOLOGICAL IDENTITY OF A CELL. ACTIVE AND REPRESSED GENES WERE DEMARCATED BY HISTONE TAIL POSTTRANSLATIONAL MODIFICATIONS EDITED BY FAMILIES OF ENZYMES KNOWN AS EPIGENETICAL FACTORS. HERE WE IDENTIFIED AND CHARACTERIZED A NOVEL METHYL-TRANSFERASE METTL8, A MEMBER OF A METHYLTRANSFERASE-LIKE PROTEIN FAMILY, TO BE SUMOYLATION DEPENDENT METHYLTRANSFERASE. SUMOYLATION OF METTL8 AT ITS N-TERMINAL EVOKES ITS LATENT CYTOPLASMIC FORM TO TRANSLOCATE INTO NUCLEUS AND BECOME ACTIVE SUMO-METTL8 FORM WHICH TRANSFERS METHYL GROUP TO HISTONE H4 IN THE NUCLEUS. THIS WOULD RESULT IN GLOBAL CHROMATIN COMPACTION AND SELECTIVE GENE ACTIVATION. ADDITIONALLY, USING CHROMATOGRAPHY PURIFICATION TECHNIQUES, WE CHARACTERIZED METTL8 TO BE A MEGA-DALTON NUCLEAR PROTEIN COMPLEX BINDING WITH TREX, AN RNA TRANSCRIPTION-EXPORT MEGA COMPLEXES AND DNA DAMAGE RESPONSE ELEMENTS TO FACILITATE CHRO
