Abstract: Stroke is currently the second leading cause of death worldwide. The quest for understanding pathophysiological roles for stroke and looking for effective stroke treatments remains an urgent priority. Compounds with a multimodal mode of action will be more preferable. In the current study, we investigated the potential role of cystathionine ?-synthase (CBS), 5-lipoxygenase (5-LO) and leukotriene B4 (LTB4) and modulator of apoptosis (MOAP-1) in the pathology of acute ischemic injury. Furthermore, we examined a pharmacological selective NF-kB inhibitor, namely andrographolide as a treatment for acute ischemic stroke and found improved stroke outcome both histological and functionally in experimental stroke animals. Our results also showed that CBS, LTB4 and MOAP-1 attributed to acute ischemic stroke pathology and inhibition of CBS improved stroke outcome. Taken together, we concluded that these systems have the potential to yield clinically effective treatment in acute stroke.
