dc.description.abstract | A sensitive and rapid assay principle for sulphite was developed based on the suppression of luminol chemiluminescence by sulphite. EDTA greatly amplified this supression. Flow-through analysis was applied to evaluate the analytical potential for measuring sulphite concentration, 50 μM H2O2, 0.5 mM luminol, 1 mM EDTA and 4 U ml-1 horseradish peroxidase being used to induce chemiluminesce. The signal of the suppression of light emission could be obtained within 1 min and was linearly proportional to the sulphite concentration from 10 to 800 μM with a relative standard deviation of < 2% over the measurable range (n = 4). Flow-injection analysis with a gas diffusion system based on this principle was applied to measure sulphite concentration (free and total) in wine. The results (free and total sulphite concentrations) can be obtained within 10 min for each wine. The results measured by the flow-injection method and an established method were compared. The assay was fairly specif toward sulphite. © 1992. | |
